Mounting medium

To make 1 mL of mounting medium, weigh out 0.9 g glycerol into a microfuge tube. Add 100 µL of a 1 M sodium borate tock, pH 9. Vortex to mix. Add 30 µg (3% by mass) N-propyll gallate and vortex thoroughly. Store in the dark at room temperature overnight to allow NPG to dissolve. Mountant is stable for a few weeks at room temperature if stored in the dark.

Paraformaldehyde

To make 100 mL of a 4% solution of fixative, agg 4g paraformaldehyde to 50 mL de-ionized water. Heat on a stirring plate until the solution reaches about 70°C. Do not allow to boil. Continue to stir and add 10 N NaOH dropwise until the solution clears. Remove from heat and allow to cool. Add remaining buffer components (i.e. PBS from concentrated stock solution) and bring the final volume to 100 mL. Adjust the pH and if the solution is still cloudy, then filter it. Store the fixative at room temperature (NOT in the refrigerator) in the dark. The paraformaldehyde solution will be stable for approximately one month.
